TIM-3 has multiple ligands, each with distinct biology:
- Galectin-9: Binds glycan structures, induces apoptosis
- CEACAM1Loading...: Protein-protein interaction, promotes exhaustion
- Phosphatidylserine: "Eat me" signal on dying cells
- HMGB1: Danger signal, blocks nucleic acid sensing
This ligand diversity makes TIM-3 biology complex and context-dependent. Functional measurement of specific ligand engagement could clarify therapeutic targeting.
