Galectin-9

gal-ek-tin nine

A TIM-3 ligand that induces T cell death, contributing to immune suppression in the tumor microenvironment.

Definition
Galectin-9 is a β-galactoside-binding lectin that functions as a ligand for TIM-3Loading.... Galectin-9/TIM-3 engagement induces T cell apoptosis and contributes to tumor immune evasion. Expressed by tumor cells, myeloid cells, and regulatory T cells, galectin-9 represents an important component of the suppressive tumor microenvironmentLoading....

TIM-3 Ligand Network

TIM-3 has multiple ligands, each with distinct biology:

  • Galectin-9: Binds glycan structures, induces apoptosis
  • CEACAM1Loading...: Protein-protein interaction, promotes exhaustion
  • Phosphatidylserine: "Eat me" signal on dying cells
  • HMGB1: Danger signal, blocks nucleic acid sensing

This ligand diversity makes TIM-3 biology complex and context-dependent. Functional measurement of specific ligand engagement could clarify therapeutic targeting.

Simplified

Multiple Partners: TIM-3 has several different ligands, not just galectin-9. Each does something slightly different. This makes TIM-3 biology complicated.

Detection Considerations

Measuring galectin-9/TIM-3 engagement requires careful consideration:

  • Galectin-9 binds via carbohydrate recognition, not direct protein contact
  • Expression levels don't indicate whether engagement is occurring
  • The interaction may be transient or lead to rapid T cell death

iFRETLoading...-based detection of galectin-9/TIM-3 proximity could potentially identify patients with active engagement of this checkpoint axis.

Simplified

Measurement Challenge: The presence of galectin-9 and TIM-3 doesn't mean they're interacting. Functional measurement of engagement would be more informative than expression alone.

Clinical Relevance

  • Prognostic value: High galectin-9 expression associated with poor outcomes in multiple cancers
  • Therapeutic target: Anti-galectin-9 antibodies in development
  • Combination potential: May enhance anti-PD-1/anti-TIM-3 combinations
  • Biomarker potential: Galectin-9/TIM-3 engagement status could guide therapy
