Viral Tracer Visualization

Mapping neural circuits with fluorescent viral reporters

Definition
The imaging of fluorescent reporter proteins expressed from viral vectors such as AAV or rabies virus to map neural connectivity, injection spread, and transduction efficiency across serial brain sections.
Anterograde and Retrograde
Different viral tools trace different directions
Quantitative Connectomics
Count labeled neurons per brain region

Circuit Mapping Workflow

Viral tracers are the primary tool for mapping neural connectivity. The workflow:

  1. Injection — stereotaxic delivery of viral vector to a target brain region
  2. Expression — wait days to weeks for fluorescent reporter expression
  3. Sectioning — serial cryosection or vibratome slicing of the brain
  4. Scanning — PanoBrain images each section with automated fluorescence acquisition
  5. Analysis — quantify labeled cells per brain region to map projection targets
