Gates

Selecting cell populations on diagrams

View
Definition
Suppose a scattergram shows a tight cloud of points. Draw a shape around the cloud, and you have a gate. In StrataQuest, a gate selects a set of events of interest within a diagram. Events inside it change color and the gate is labeled, so the population can be followed back to the image. Dividing events into positive and negative along one measurement is the job of cutoffsLoading....

How It Works

To draw a gate, open the Gates menu from the diagram’s toolbar and choose a shape:

  • Rectangle: histograms, scattergrams, t-SNE, UMAP, SONG and phenotype diagrams.
  • Ellipse: the same diagrams except histograms.
  • Polygon.

Releasing the mouse fixes the gate. Events inside it change to a predefined color and the gate is labeled (for example, “Gate 2”); you can edit the color and the label afterward. From a gate you can open the Raw Data of its events and highlight them on the images.

Simplified

Draw a shape around a cloud of points on a plot, and those cells become a named group you can find again in the image.
